High-performance, chemically-defined kit for superior expansion of human mesenchymal stem cells (hMSC)

RocketCell™ hMSC Xeno-Free Complete Growth Medium is a fully defined, animal component–free formulation designed for robust, rapid, and reproducible expansion of human mesenchymal stem cells (hMSCs). This complete medium system integrates an optimized basal medium with a chemically defined supplement to ensure high stability and potency for consistent, long-term performance.​

The kit has been rigorously validated with multiple mesenchymal stem cell lines, producing uniform cultures with key multipotency markers, including CD73, CD90, and CD105. For smooth transition from serum-based media, initial culture on fibronectin-coated plates is recommended. After short-term adaptation, cells can be expanded on standard cultureware without additional coating.​

CASE 1

Growth of hMSCs Cryopreserved for 20 years in
​RocketCell™ hMSC Xeno-Free Complete Growth Medium. ​

An ampoule of hMSCs was defrosted after 20 years of storage in liquid nitrogen vapor. Cells were plated into a 24-well fibronectin-coated VitroPrime™ Spread Attach plate and placed into a low O2 incubator (6% O2). The media was changed after 48h, and at 72h, the plate was placed in an IncuCyte S3 and scanned every 4 hours to track cell growth. ​

hMSC cultures grown in a Fibronectin-coated T75 flask were passaged using TrypLe and plated onto a Fibronectin-coated 24-well VitroPrime™ Spread-Attach Plate at a density of 10,000 cells/well (~5000 cells/cm2). The plate was immediately placed into an Incucyte S3 and imaged over thecourse of 163h, with media changed every 48 hours. ​

​hMSC cultures were passaged from T75 Fibronectin-coated flask and plated on
uncoated, Vitronectin (10 ug/mL) or Fibronectin (10 ug/mL) coated 12-well VitroPrime™ Spread-Attach Plate. The cells were incubated for 72 hours, then imaged on an Incucyte S3.

Immunostaining of hMSCs Cultured in​ RocketCell™ hMSC Xeno-Free Complete Growth Medium​​

hMSC cultures were plated on Fibronectin (10 ug/mL) coated 24-well VitroPrime™ Spread Attach Plate. After 6 days, the cells were processed for standard immunofluorescence and stained with directly labeled antibodies: CD90-FITC, CD105-PE, and CD73-APC. The staining was observed and documented using a Leica MICA confocal microscope.

Tri-Lineage Differentiation of hMSCs Cultured in
RocketCell™ hMSC Xeno-Free Complete Growth Medium​​

hMSC cultures were plated on a 24-well Fibronectin (10 ug/mL)- coated VitroPrime™ Spread Attach Plate. After 6 days of expansion in RocketCell™ hMSC Xeno-Free Complete Growth Medium, the cells were placed into Adipogenic, Chondrogenic, and Osteogenic Media for 12 days. The cultures were fixed and processed for standard chromogenic staining with Oil Red O for adipocytes, Alcian Blue for chondrocytes/cartilage, and Alizarin Red for Osteocytes. The staining was observed and documented using a Keyence BZX microscope.
