Background:
- Organoids are 3D self-organizing tissue models that closely mimic the architecture and function of native organs.
- Epithelial polarity is a fundamental feature of intestinal organoids, governing nutrient absorption, drug transport, and barrier function.
- Conventional Matrigel-grown intestinal organoids exhibit an apical-in orientation, restricting direct access to the absorptive epithelial surface.
- Apical-out intestinal organoids expose the brush border to the external environment, enabling direct interaction with nutrients, drugs, microbes, and
other luminal factors. - Apical-out polarity improves the physiological relevance of in vitro models for studying intestinal biology, nutrient uptake, and oral drug absorption.
- Most current organoid culture systems rely on animal-derived extracellular matrices, which introduce batch-to-batch variability and limit translational
applications. - Synthetic, xeno-free hydrogels provide a chemically defined and reproducible microenvironment for organoid culture, improving experimental consistency
and scalability. - Developing robust apical-out organoid models in synthetic hydrogels can enhance preclinical research in nutrition, drug discovery, disease modeling, and host–microbiome interactions.
